Abstract:
Inflammation is a key protective response and most of the mechanisms that link inflammation to damage repair and regeneration are conserved within evolution. Inflammation is a finely tuned response; perturbation of this response leads to many chronic and degenerative diseases. Development of new strategies to treat inflammation and resulting pathologies is urgently needed, in view of the long long-term effectiveness of existing treatments. The current research proposal concerns major problems of the public health, namely chronic inflammatory diseases and associated diseases. Inflammatory Bowel Disease (IBD) is a group of inflammatory pathologies affecting the digestive tract that may evolve in extremely disabling cancers. Ulcerative Colitis (UC) and Crohn’s Disease (CD) are the main types of IBD. Currently, 3 million people are identified worldwide as patients of IBD. The incidence of IBD has increased tenfold in the last 15 years in Romania. These pathologies, which primarily affect young adults (20-30 years), are as frequent as insulin-dependent diabetes, and 3 times more common than multiple sclerosis. This project, based on a multidisciplinary approach should have a profound impact on our understanding of the role of P2RX7-dependent signaling in inflammation and cancer and should contribute to the discovery of a successor of the 5-ASA which is the basic and symptomatic treatment of IBD.

Abstract:
After cardiovascular diseases, cancer is the second cause of mortality in the world. Thus, it is necessary to study this disease and discover new anticancer drugs. In this light, our main objectives are to develop new compounds with anticancer potential, fit the nature of the substituents onto the starting materials in certain cases in order to reinforce the metabolic stability of final compounds, to evaluate the anticancer potential of synthesized compounds on tubulin polymerization, on human farnesyltransferase and on cancer cell lines and to establish structure-activity relationships for resulting anticancer agents. The used strategy will be one of the improvement of the biological activity, by drug-design methods, of 'hits' with anticancer potencies (IC50 in the 10-8 molar range), resulting from a previous research work realized by the project leader Pharmacomodulations will be realized on the phenothiazine ring, on the connector and on the benzenic/indole ring via modern synthetic organic methods.
