Abstract:
Wound care management represents a major health care problem in the entire world and biomedical research is constantly seeking for new options with respect to promoting the healing process and to reduce the therapy cost. The goal of the project is the design, development and evaluation of new biomimetic and bioactive polymeric nanostructures (BBPNs), for the treatment of acute and chronic wounds. The innovative designed nanostructures are based on chitosan/chitosan-hyaluronic acid nanoscaffolds, which have incorporated curcumin, propolis, honey, insulin, L-arginine or allantoin, as active substances, with important biological effects such as antimicrobial, antioxidant anti-inflammatory and healing. The innovative BBPNs will be: (i) characterized using Scanning Electron Microscopy, Transmission Electron Microscopy, FT-IR spectroscopy, X-Ray Diffraction, Thermal behavior; (ii) studied in terms of physico-chemical and mechanical parameters (porosity and swelling degree, contact angle measurements, tensile strength, entrapment efficiency, rheology, water vapor transmission); (iii) kinetic studies of delivery of the active substances; (iv) in vitro assays (antimicrobial and antioxidant effects, citotoxicity and hemocompatibility, cell migration and wound closure); (v) in vivo assays (wound model induced to diabetic and nondiabetic rats). The novelty of the project is supported by the original design of the BBPNs which assure antimicrobial, anti-inflammatory and healing effects

Abstract:
The contamination and infection of the burn wound, and especially the sepsis, by delaying the healing process, remain the most serious complications in the burn trauma management. The goal of the project is the preclinical evaluation of innovative therapies for the topical treatment of burn wounds in which an antimicrobial agent is associated with a promoter of healing in original formulations. Through innovative and original approach new sulfonamides that have attached structures with known antimicrobial (azetidin-2-one) and healing (arginine, allantoine) properties will be designed. After synthesis and confirmation of the structure through spectral methods (UV, FT-IR, 1H-NMR, MS) the toxicity degree, the antimicrobial (on Gram positive and Gram negative bacteria strains) and healing (on animal burn wound model) tests will be performed. Simultaneously hydrogels based on chitosan and hyaluronic acid will be developed and their chemical, pharmacokinetic and pharmacological characteristics will be established. Finally, the most active designed compounds will be included into biopolymer hydrogels in original formulations. These new therapeutically systems will be studied in terms of the kinetic parameters of topical delivery of the active substances and of the healing potential. We expect that this new innovative formulations will solve both the problem of bacterial infection and will also hasten the healing process favoring rapid full coverage and the recovery of the patient.

Abstract:
Although mitochondrial disorders are associated with different type of pain, the role of mitochondrion in pain processing has been far less studied until recently. In the last decade there are more and more evidences that mitochondria plays an important role in pain processing by modulating radical oxide species production as well as ATP and intracellular calcium control.
The project main aim is to determine the mitochondria involvement in pain processing and modulation by looking at the link between the metabolic rupture of mitochondria homeostasis (systemically and in the cortex) and the activation of spinal neurons in different models of acute, chronic and visceral pain.
By administrating antagonist drugs that use the same mitochondrial pathways to stimulate and inhibit the mitochondrion we will analyze the pain behavior modulation and the release of different substances (H+, ROS, HIF) on the cortical surface and cortico-brain stem/spinal cord pathways.
In these studies, we will use state-of-the-art neuroscience techniques, including neuronal tracing, immunocytochemistry and western blots, as well as behavioral and pharmacological approaches that allow us to pinpoint the brain regions of interest and to identify specific neural mechanisms.
The results of our studies will allow a better understanding of the role of mitochondria and free radicals in different types of pain and find future approaches in pain treatment.
