Abstract:
The present project proposal is part of the research theme related to the synthesis, characterization and biomedical applications of intelligent magnetic hydrogels, based on magnetic nanoparticles, loaded with triterpenic antitumor active agents (betulinic and ursolic acids). Within the project, magnetic nanoparticles of iron oxides (Fe3O4 and γ-Fe2O3) will be synthesized using green synthesis (extracts of Taxus baccata L. and Trametes versicolor L.), followed by their encapsulation together with the antitumor agent in a nanostructure of hydrogel type. The magnetic hydrogel will have applicability in cancer, through thermal treatment, the so-called magnetic hyperthermia. The element of originality consists in the development of hydrogels that will present a synergistic antitumor behavior, with improved pharmacological properties. The compatibility of the hydrogels with the human body will be performed on healthy human cell lines (alveolar epithelial cells and human hepatocytes), and the antitumor effect will be investigated using human tumor cell lines (lung cancer, hepatocellular carcinoma). The biosafety profile of the hydrogels will be evaluated using 3D functional microtissues from normal bronchial cells (EpiAirwayTM) and primary human alveolar epithelial cells (EpiAlveolarTM). These functional 3D microtissues constitute in vitro model systems developed to mimic biological responses relevant to humans.

Abstract:
The experience gained during over ten years of research in the field of natural compounds lead to the identification of a promising natural candidate for a skin problem with which, according to statistics, about half of the population is facing. This problem refers to skin tags (acrochordons), common fibroepithelial benign lesions, with a high rate of prevalence in the adults public, and strongly associated with obesity, insulin resistance and metabolic syndrome. Acrochordons are currently eliminated most commonly by electrocautery, surgical excision or freezing. However among dermatologists it is also recognized that oregano essential oil (OEO) can, in a non-invasive manner eliminate skin tags. Up to date there are no scientific proofs regarding the mechanism of action neither a well established carrier designed in order to eliminate the possible caustic effects (it is generally accepted that essential oils should not be applied directly on the skin ). The aim of this project is to provide and characterize an advanced pharmaceutical formulation for the incorporation of OEO type innovative hydrogels containing polymeric micelles as drug delivery systems (OEO-PMs) as well as to characterize the vitro, in ovo and in vivo effects. The screening will be forwarded towards the assessment of the effect of OEO-PMs hydrogel on the main components of skin tags (keratinocytes, blood vessel- angiogenesis) and also towards the immunomodulatory potential. Modifications in skin characteristics after application (transdermal water loss, erythema, melanin, histological changes) will be analyzed as well as lack of adverse reactions. A success in these directions will lead to both an innovative research line as well as to the possibility of transfer of knowledge to the industry and last but not least to solving the problem of skin tags in a well documented non-invasive manner.

Abstract:
Anthocyanins, natural plants pigments found in very significant amount in edible berries and many vegetables, showed to have high antioxidant activity. Because of this they are featured as primary ingredients in food products, natural supplements and cosmetic industry. Multiple in vitro studies showed that they have antiproliferative, anticarcinogenic and proapoptotic effects. It is also known that they are very susceptible to degradation and their stability is highly influenced by pH, temperature, light and the presence of other phenolic acids.
Unfortunately it is not clear and was not investigated, how exactly anthocyanins act at cellular level and what is their real availability, cell uptake and if they can penetrate skin without degradation.
Current project propose to answer these questions and provide a better understanding of their effects on melanoma cell lines. Its final goal is to determine the exact amount of anthocyanins uptaken by the cell. This information is crucial for pharmaceutical industry in order to develop recommendations for topical application and using anthocyanins as chemopreventive ingredients. Melanoma was chosen because it is responsible for approximately 75 % of all deaths from skin cancers. Considering this, the proposed project brings a new approach to a very common problem in Biotechnology today.

Abstract:
The aging phenomenon and the aging itself are considered major challenges in cosmetology and dermatology research and the use of vegetable principles in combination with nanotechnology represents a current topic in the formulation of therapeutic and prophylactic products.
The current proposal focuses on the synthesis and testing of a dermatocosmetic formulation based on gold nanoparticles and betulin, with anti-aging valences due to gold, considered as both therapeutic agent and delivery vehicle, as well as betulin, active agent that requires suitable delivery at skin level. The main objective, consisting in the preparation of semisolid modern formulations with gold nanoparticles and betulin, can be accomplished through the implementation of appropriate standard technologies (ISO), in addition to the practical training. The project proposes the analysis of a dermato-cosmetic product starting from the preparation and characterization up to the industrial scale production, involving the quality control of the final product.
Due to the beneficial effects at skin level of both compounds, the project proposes a thereof association in order to achieve an optimal semisolid pharmaceutical formulation in preventing and treating skin aging. The related activities are focused on: the preparation and analysis of betulin-gold nanoparticles formulations, the in vitro assessment of their toxicity and effectiveness, according to ISO 10993 and other current standards, the antimicrobial testing and testing on healthy volunteers through: consumer test, sensory evaluation, non-invasive tests.
The final goal of the proposal is the transfer of academic scientific research results towards the economic agent in order to implement a continuous flow of production and testing for modern cutaneous cosmetic formulations.

Triterpenele pentaciclice, implicit şi acizii oleanolic şi ursolic, suscită un real interes în cadrul cercetărilor privind compuşii naturali antitumorali şi antiinflamatori. Dezavantajul major prezentat de triterpene este solubilitatea lor extrem de scăzută în apă, care poate determina o biodisponibilitate redusă. Proiectul îşi propune optimizarea solubilităţii în apă a unor triterpene pentaciclice, ce se poate realiza prin complexarea cu ciclodextrine hidrofile. Complecşii ciclodextrinici se obţin prin diverse metode şi se pot analiza fizico-chimic prin metode consacrate (dizolvare in vitro, analiza termică, difracţie de raze X, difuziune in vitro, etc.). Obţinerea nanostructurilor cu ciclodextrine amfifile şi triterpene se poate realiza spontan, în anumite condiţii, şi duce la posibilitatea terapiei la ţintă cu o cantitate mai mare de substanţă activă. Complecşii obţinuţi sunt supuşi analizei in vitro, prin testul MTT pe linii celulare tumorale, linii celulare care se vor utiliza pentru producerea modelelor animale prin inocularea la şoarecii C57BL/6J şi la şoarecii atimici BALB/c. Testele in vivo constau în testul CAM, pe ouă embrionate, pentru relevarea capacităţii antiangiogenice ale substanţelor active, precum şi în teste pe modelele animale obţinute anterior. Ultima etapă este reprezentată de evaluarea histologică şi imunohistochimică. Formulările cele mai performante vor fi brevetate şi publicate.

Abstract:
Flavonoids are a class of natural molecules with multipotent biologic properties arousing a true interest in therapeutics and micronutrition for their capacity to reduce the incidence of various cancer types. Among them, aurones and chalcones are less studied subclasses, with emerging biological potential. A major drawback on the therapeutic use of flavonoids is their reduced solubility in water. The project aims at preparing nanostructures with maximal bioavailability employing a standardized plant extract and chemopreventive flavonoids. The objectives of the project are : 1) to obtain an extract enriched in aurone and chalcone type flavonoids using as prime matter the wood of Cotinus coggygria; 2) the inclusion of the extract and some active principles (sulfuretin, butein) in cyclodextrins in order to enhance their hydrosolubility; 3) to obtain bioconjugates with rapid metabolization by derivatizing active flavonoids (sulfuretin, butein) followed by their complexation with cyclodextrins; 4) physic-chemical characterization of the obtained products; 5) in vitro evaluation of the soluble complexes on normal cells and five cancer cell lines (A375, A431, MDA-MB-231, HepG2, HT29) as well as the evaluation of anti-inflammatory activity on murine macrophages RAW264.7; 6) in vivo tests on embryonated eggs (CAM test for the evaluation of anti-angiogenic capacity) and on athymic Balb/c mice inoculated with cancer cells. The most efficient formulations will be subjects for patents.
