Abstract:
Project director, associate professor, doctor of sciences, titular, of the discipline of genetics Ioan SARAC.
Hazel (Corylus avellana) belongs to the family of trees as the birch (family Betulaceae), however it is described as a bush rather than a tree, as it tends to
produce several 'trunks' or shoots rather than just one. The edible nuts grow in groups of up to four; they reach two centimetres in size and are sheathed by papery modified leaves.
The English name for this tree derives from the Anglo-Saxon 'haesel knut'; haesel means cap or hat, and refers to the papery cap of leaves on the nuts. Importance of germplasm in Corylus avellana L.
Basic concepts in the field of conservation and use of germplasm in Corylus avellana L. Evaluation of germoplasm sources in Corylus avellana L.: importance, objectives, stages.
Primary evaluation at the collection site and secondary assessment in the laboratory and experimental field. Phytosanitary quarantine measures in the circulation of germoplasm sources in Corylus avellana L. Determination of genetic diversity
by means of molecular markers at Corylus avellana L.

Abstract:
The diseases caused by the fungus belonging to the Oomycetes are producing every year very severe damages on some important horticulture crops like vineyard, potato, tomato and cucumber. There are some agronomical measures which can reduce the level of infection, but they can not decrease significantly below the Economic Injury Level. From this reason the vegetables growers and vineyards growers, has to look for combined methods, including the chemical control of these pathogen agents. Actually the most important way for controlling of these diseases, is the treatment on vegetation with different kind of fungicides realized via chemical synthesis. Due to the mistakes made in application of these products, the number of applications was increased too much and the resistant races of fungus to different active ingredients have appeared. From this reason, it is more and more difficult and more expensive to control Oomycete fungus by chemical methods. On the other hand, these products are killing many useful organisms. They prove a very serious risk for the health of the operators. Also, due to the high level of residues coming from these chemical products into the fruits harvested from treated plants, there is a high risk for the consumers too.
The main objective of the our project is to obtain a new formulas effective, with lowes negative impact on the environment, with the antifungal action on Oomycetes that attack horticultural crops and the capitalization antifungal potential of plants.
The project also stimulates partnership between a research institute, two SMEs and a NGO, and helps to ease the road from scientific knowledge to technology with socio-economic impact, in line with market needs and evolution.

Abstract:
Cadmium has been classified as a group 1 human carcinogen: soluble Cd salts accumulate and result in toxicity, causing a variety of adverse health effects. Metallothioneins are classified as Cd-binding proteins, they are involved in detoxification of heavy metals, and scavenging of free radicals. Concentration dependent expressions of metallothionein genes were measured by reverse transcription followed polymerase chain reaction in the brain, heart, muscle, kidney and liver of control and cadmium (1 mg/l or 10 mg/l Cd2+) treated animals. Both the MT-1 and MT-2 mRNA levels increased with Cd concentration in a time- tissue- and dose-dependent manner. The expression of MT-2, however, was always more responsive to a high Cd concentration. In parallel with the induction of the MTs by Cd, we followed the accumulation of this metal in the kidney and liver. Although the Cd level was always higher in the kidney during treatment, the rate of accumulation was higher in the liver. We analyzed the in vivo effects of cadmium ion on the hepatic detoxifying cytochrome P450 enzyme system of common carp (Cyprinus carpio L.) After treatments we measured the P450 content, EROD and ECOD isoenzymes activities in the liver microsomes. We have demonstrated that the heavy metals induce the biotransformation enzymes in low concentration, but at the high ion concentrations enzyme inhibition was observed. The digestive enzymes in the gastrointestinal tract can be directly exposed to the different xenobiotics. The aim of experiments was to study the effects of different heavy metals on the activities of some digestive enzymes:trypsin, α-chymotrypsin, carboxypeptidase A and lipase. In the in vitro experiments we firstly investigated the effect of different concentration of Cd on activities of these enzymes. The enzymes were isolated from common carp (Cyprinus carpio L.) and partially purified for the measurements. Slightly influenced the presence of Cd ions the activites of proteolytic enzymes. 20% decrease was measured in lipase activity. As transtion metal ions have special role in generation of oxygen radicals, we investigated the antioxidant defense system of fishes. Reactive oxygen species can cause oxidative stress in absence of effective antioxidant system. Signs of oxidative stress (lipid peroxidation) and depletion of reduced glutathione (GSH) were tested. Our results show increasing lipid peroxidation and decreasing reduced glutathione levels. Changes in antioxidant enzyme activities (SOD, C-ase, GPx) refer to oxidative predominance also.

Abstract:

Abstract:
The project aims to identify plant species susceptible to the presence of heavy metals in large quantities in the soil and to use these plants as a quick tool for assessing soil quality by early detection of possible functional changes and the component of biological systems, but also aims to study the reaction and response of the bio-systems to environmental changes. The proposed study will attempt to assess the quality of soil by testing different species of plants that have selectivity for heavy metals but also the use of these plants in therapeutics. A unified and complete documentary study on bio-indicating plants and their use for cosmetic and pharmaceutical purposes will be prepared. This documentation will be permanently updated in relation to the evolution of the research theme. The proposed project will contribute to the development of a university research department of major importance for environmental protection. The novelty of the project consists in studying the degree of soil pollution by two methods: an active biomonitoring using species introduced into the environment (sentinel species), which reacts relatively quickly to the increase in the concentration of heavy metal and the other biomonitoring passages with the help of some detector species, which grow naturally in a certain area and have a slow reaction to increasing pollutant concentration. As a research paper, the study will broaden the knowledge base on the influence of heavy metals on the development of some plant species. The end result of research is the use of plants identified as heavy metals indicator in cosmetics and dermatology. Extracts of these plants enriched in metals favor dermal retention of water and can be used in ionic exchange for hydration because it favors the dermal recovery effect.
